When striatal homogenates were used for these studies, a single, high-affinity binding site was found. In vivo studies revealed that spiperone bound specifically to the striatal complex. The ratio of spiperone binding in the striatum to that in the cerebellum was significantly higher than that in vivo; the latter value represents the ratio expected by simple brain distribution. Saturation studies in striatal homogenates indicate that [3H]spiperone binding is competitively inhibited by spiperone and spiroperidol, but not by fluphenazine, chlorpromazine, imipramine, zotepine, nemonapride, clozapine or risperidone. In vitro pretreatment of striatal homogenates with 0.1 microM spiperone reduced the number of spiperone binding sites to about half the number found in controls; the binding sites were found to be of a single affinity.
